Deletion of TOR or SCH9 increase life span additively with deletion of FOB1 but not with dietary restriction. These data support the model that TOR, SCH9 and dietary restriction act in a single pathway that is distinct from the pathway that FOB1 acts in. Deletion of FOB1 is believed to increase replicative life span by reducing the accumulation of extrachromosomal rDNA circles (ERCs) in the mother cell [10207108]. |

Deletion of both GPA2 and FOB1 significantly increased life span in the BY4742 background. The double deletion mutant was longer-lived than either single mutant consistent with the model that FOB1 and GPA2 act in different pathways to modulate replicative life span in yeast. |

Addition of 5 mM nicotinamide has no effect on replicative life span in BY4742 sir2 fob1 double mutant cells. Dietary restriction is still able to increase life span of sir2 fob1 double mutant cells treated with 5 mM nicotinamide. These data demonstrate that SIR2 and other nicotinamide-sensitive sirtuins are not required for life span extension from dietary restriction in the BY4742 strain background. |

Deletion of TOR or SCH9 increase life span of sir2 fob1 double mutant cells. SIR2 and FOB1 are believed to act in a single genetic pathway to promote replicative life span by reducing the accumulation of extrachromosomal rDNA circles in the mother cell [ 10521401]. Since dietary restriction also increases the life span of sir2 fob1 double mutant cells [ 15328540], this supports the model that TOR1, SCH9, and dietary restriction act in a single pathway that is distinct from SIR2, FOB1, and extrachromosomal rDNA circles. |

Dietary restriction at 0.5%, 0.05%, or 0.005% glucose significantly increases the replicative life span of sir2 fob1 hst1 hst2 quadruple mutant cells in the BY4742 strain background. |

Deletion of HXK2 significantly increases the replicative life span of sir2 fob1 hst1 hst2 quadruple mutant cells in the BY4742 strain background. These data demonstrate that SIR2, HST1, and HST2 are dispensable, both individually and in combination, for life span extension from deletion of HXK2. Deletion of HXK2 is proposed as a genetic mimic of dietary restriction [ 11000115]. |
